Our working group deals with the process of intramembrane proteolysis, which combines protein degradation with signal transduction and thus represents an important regulatory mechanism for cellular homeostasis. We are currently particularly interested in the SPP/SPPL family of intramembrane proteases, which include the signal peptide peptide (SPP) and various SPP-like homologs. These proteases cleave specific membrane proteins and thus contribute to the homeostasis of the cellular membrane proteins while simultaneously acting as regulatory switches. In recent years, we have extensively investigated the function of these enzymes, both using cell-based approaches and mice as model organisms. Based on this work, we were able to identify roles of SPP/SPPL proteases in the immune system, arteriosclerosis, germ cell development and regulation of glucose metabolism. In the planned PhD project, we would like to further characterize the substrate spectrum and the in vivo function of SPP. The project will therefore combine basic biochemical/cellbiological experimental approaches with the phenotyping of conditional knockout mice, in particular with regard to metabolism and the immune system. The project includes a wide range of experimental techniques. The question combines basic biochemical/cellological questions with the characterization of pathophysiological relevance of biochemical findings and their translational potential.
